Conditionally Replicating Vaccine Using Suppressor tRNA Technology Protects Pigs Against Seneca Valley Virus

Researchers have developed a conditionally replicating live vaccine that protects pigs against the Seneca Valley virus (SVV). The study, published in *npj Vaccines* on June 23, 2026, demonstrates that the vaccine utilizes suppressor tRNA technology to trigger an immune response in swine while preventing uncontrolled viral replication.

The research team engineered the vaccine by incorporating an amber stop codon into the viral genome, which requires the presence of an exogenous suppressor tRNA for the virus to replicate. In the laboratory setting, the vaccine successfully replicated in cells expressing the suppressor tRNA but remained inactive in standard host cells. When researchers administered the vaccine to pigs, the animals developed high levels of neutralizing antibodies and showed no clinical signs of disease after a subsequent challenge with the wild-type Seneca Valley virus. The findings indicate that this platform provides a stable method for immunization, as the vaccine maintains its safety profile while effectively priming the immune system against future infection.
