December 25, 1982: The DNA Sequence of Bacteriophage λ Has Been Determined

On December 25, 1982, the American biochemist Frederick Sanger completed the sequencing of the bacteriophage λ using dideoxy chain termination method in conjunction with random cloning in M13 vector. Sanger not only found that the circular DNA contained 48,502 base-pairs, but also clearly identified the reading frame for its 46 genes. The sequencing of bacteriophage λ is of great significance for elucidating the principle of gene recombination and the development of genetic engineering.
