Study Identifies PHIP Protein’s Role in Inhibiting NuRD Complex to Promote Cancer Growth

A recent study published in *Nature Communications* has identified a significant molecular interaction that may play a role in driving cancer growth. Researchers, led by Malone and colleagues, have discovered that PHIP, a protein involved in cellular processes, inhibits the NuRD complex—a key chromatin remodeling mechanism—thereby enhancing the activity of the SWI/SNF complex. This interaction appears to contribute to the unchecked cell proliferation observed in certain cancers.

Chromatin remodeling complexes like NuRD and SWI/SNF regulate gene expression by altering the structure of chromatin, which is critical for maintaining normal cellular function. The study highlights how PHIP disrupts NuRD’s regulatory functions, effectively tipping the balance in favor of SWI/SNF activity. This imbalance may create conditions conducive to tumor growth.
